A RP-HPLC method for the simultaneous estimation of Silymarin and Thiamine in bulk and pharmaceutical forms

ABSTRACT:
For the simultaneous estimation of Silymarin and Thiamine in bulk and pharmaceutical dosage forms a rapid, simple reverse phase high-performance liquid chromatographic method was developed and validated. Using Inertsil ODS C-18, 5┬Ám column with 250 x 4.6mm internal diameter, chromatographic analysis was carried out with a mixture of methanol: acetonitrile: tetrahydrofuran in the ratio of 30:65:05 (v/v/v) as mobile phase. Different analytical parameters such as linearity, precision, accuracy, specificity, limit of detection (LOD) and limit of quantification (LOQ) were determined. The calibration curve was found to be linear for each analyte in the desired concentration range. The percentage recovery was found to be 101.17 and 99.86 for Silymarin and Thiamine respectively. It is substantiated from the LOD value of 0.5 and 0.1 ppm for Silymarin and Thiamine respectively that the proposed method is highly accurate, sensitive and precise. Hence the present method can be applied successfully for the quantification of active pharmaceutical ingredient (API) content in the combined formulations of Silymarin and Thiamine.
