ABSTRACT:
A simple, selective, accurate Reverse Phase High Performance Liquid Chromatographic method was developed and validated for the simultaneous estimation of Moxifloxacin hydrochloride and Cefixime trihydrate in their synthetic mixture. Chromatographic separation achieved isocratically on a thermoscientific BDS Hypersil C18 (250mm X 4.6 mm i.d., 5 µm particle size) column utilizing a mobile phase of acetonitrile: tetrabutyl ammonium hydrogen sulphate (TBAHS) + tetrabutyl ammonium bromide (TBAB) buffer pH 3 (20:80 v/v) and adjusting pH 3 with orthophosphoric acid at a flow rate of 1.5ml/min with UV detection at 280.8nm. The retention time for Moxifloxacin hydrochloride and Cefixime trihydrate was found to be 3.078 and 5.263 min respectively. Linearity was obtained in the concentration range of 10-60µg/ml of Moxifloxacin hydrochloride and 10-60µg/ml of Cefixime trihydrate with a correlation coefficient of 0.999 and 0.999 respectively. The developed method was validated in terms of accuracy, precision, linearity, limit of detection, limit of quantitation. This study aimed at developing and validating an HPLC method being simple, accurate, robust and selective and the proposed method can be used for the estimation of these drugs in combined synthetic mixture.
