ABSTRACT:
Vitex negundo L. (Nirgundi) is traditionally used in Ayurveda and folk medicine for pain, inflammation, swelling, arthritis, wounds and other inflammatory disorders. Its therapeutic potential may be due to the presence of flavonoids, phenolics, terpenoids, glycosides and alkaloids. The present study has been aimed to evaluate the phytochemical profile of ethanolic extract of Nirgundi seeds using TLC, UV-Visible/IR spectrophotometry and FT-NMR Spectroscopic techniques, and thereby evaluate their potential role in plant-authentication. TLC showed multiple fluorescent bands, indicating the presence of different phytoconstituents especially phenolics and flavonoid compounds. Further, UV-Visible spectrophotometry highlighted strong absorption profile at 220-350 nm suggesting the presence of conjugated systems as seen in flavonoids and phenolics. FT-IR analysis has indicated the presence of functional groups such as -OH, -CH, -C=O, and aromatic -C=C- (ring double-bonds). The highlight of the present study lies in the involvement of 1H- and 13C NMR spectral information to identify few phytoconstitutents present in the seed-extract of V. negundo, which could be used as finger-print markers in plant-authentication. FT-NMR Spectroscopic analysis clearly indicated the presence of aromatic-, olefinic-, hydroxyl-, allylic- and aliphatic functionalities, confirming the presence of flavonoids and triterpenoids-type compounds such as Luteolin, Casticin and Ursolic acid. NMR spectral analysis has further indicated H-bonding interaction between a ring –OH and an adjacent ring –C=O group, possibly belonging to either Luteolin or Casticin. The current study underscores a preliminary chromatographic and Spectroscopic analysis of bioactive phytoconstituents in the ethanolic extract of Vitex negundo L., which could be used as an additional method for plant-authentication beside the conventional methods.
